Spike protein is an envelope-anchored protein that mediates the recognition and binding of coronaviruses (SARS-CoV-2/COVID-19) to host cells. Spike protein can be further cleaved by the host protease into two subunits called S1 and S2, wherein the S1 subunit contains a receptor binding domain (RBD) crucial for the specific recognition and interaction with the host cell receptor, angiotensin converting enzyme-2 (ACE2). This is the first and the most essential step for the virus infection. S1RBD is the main target of neutralizing antibodies upon infection. Drugs/compounds that inhibit formation of the S1RBD-ACE2 complex in vitro could be potential treatment for COVID-19 infection.
SARS-CoV-2 S1RBD-ACE2 Binding Assay Kit is designed to measure the interaction between S1RBD and ACE2. The assay can be used to detect neutralizing antibodies in human plasma or serum, and to screen for inhibitors and potential drugs which inhibit the S1RBD-ACE2 interaction. It can also be used for vaccine development.
